一株抗马铃薯晚疫病菌的粘细菌鉴定及其活性分析

    摘要:

    本文采用大肠杆菌诱导的方法,从内蒙古呼和浩特市采集的土壤中分离到一株细菌(Xt-2)。通过子实体和菌落形态学的初步观察、生理生化特征和16S rDNA分子鉴定,该菌株具有与橙色粘球菌相似的形态及生理生化特征,且其16S rDNA序列与橙色粘球菌的16S rDNA序列具有99%的同源性,说明该菌株为橙色粘球菌。抗菌活性表明菌株Xt-2具有明显的抗马铃薯晚疫病菌活性,且活性物质主要存在于菌株的胞外代谢产物中。活性物质不是蛋白质,对紫外的耐受性比较好,但对高温、过酸或过碱的环境比较敏感。本研究为菌株Xt-2抗马铃薯晚疫病活性物质的提取、分离及鉴定奠定了基础,也为内蒙古地区粘细菌的深入研究及抗马铃薯晚疫病新药研发奠定了一定的基础。

    Abstract:

    In this study, a strain Xt-2 was isolated from the soil samples by E. coli induction method in Hohhot, Inner Mongolia. The strain had the similar morphological, physiological and biochemical characteristics as Myxococcus fulvus and its 16S rDNA sequence shared 99% homology with that of Myxococcus fulvus, suggesting that the strain Xt-2 belonged to Myxococcus fulvus. The analysis of antibiotic activity showed that the strain had a high resistance against phytophthora infestans and the active substances mainly existed in the extracellular metabolites of the strain. The active substances were not protein and tolerant to UV light, but sensitive to the high temperature, too acidic or too basic environment. This study laid a foundation for the extraction, separation and identification of the active substances resistant to phytophthora infestans and the further research on myxobacteria and the discovery and development of the new drugs against phytophthora infestans in Inner Mongolia area.
