      摘要 目的 研究木犀草素对血管紧张素Ⅱ诱导乳鼠心肌细胞肥大的作用。方法 用1到3天新生大鼠分离心肌细胞;用徕卡倒置显微镜抓获心肌细胞图像以测量细胞直径;用BCA蛋白检测法测定心肌细胞蛋白质含量;以[3H]苯丙氨酸掺入法测定心肌细胞的蛋白质合成率。结果 0.1 μM血管紧张素Ⅱ引起了心肌细胞直径、蛋白含量和心肌细胞蛋白质合成速率的显着增加。木犀草素预处理可剂量依赖性的减小由Ang II刺激而引起的乳鼠心肌细胞直径、蛋白质含量和蛋白质的合成速率增加。结论 结果表明,木犀草素可有效抑制血管紧张素Ⅱ诱导的心肌细胞肥大。

      [ABSTRACT] AIM:To examine the effect of Luteolin on angiotensin II-induced hypertrophy of cultured neonatal rat myocardial cells. METHODS:Neonatal cardiomyocytes were prepared from the ventricles of 1 to 3-day-old rats. To determine cell diameter, myocardial cell images captured under a Leica inverted microscope were measured. The protein content was measured using a BCA protein assay. As an index of protein synthesis, the rate of protein synthesis of myocardial cells was measured by incorporation of [3H] phenylalanine. RESULTS: 0.1 μM Angiotensin II treatment alone caused significant increases in the diameter, protein content and the rate of protein synthesis of myocardial cells. Pretreatment with Luteolin significantly attenuated cell diameter, protein content, and the rate of protein synthesis in neonatal cardiomyocytes stimulated with Angiotensin II in a dose-dependent manner. CONCLUTION:Our results indicate that Luteolin is an effective inhibitor for angiotensin II-induced cardiomyocyte hypertrophy.
